Core Concept: Human artificial chromosomes offer insights, therapeutic possibilities, and challenges.

چکیده
Francesco Saverio Tedesco works with a big gene. In fact, the human dystrophin gene, with its whopping 2.4 million nucleotides, is one of the largest found so far in nature. A clinician-scientist at University College London, Tedesco hopes to use gene therapy to replace a faulty version of dystrophin in people with Duchenne muscular dystrophy. But the large deletions in some patients are too big to fix with gene editing, and the entire dystrophin gene is too large to fit inside the viruses normally used to deliver replacement DNA. Instead, Tedesco and his colleagues turned to human artificial chromosomes, or HACs, which can hold an essentially unlimited amount of DNA. They have already used HAC delivery to improve movement and other muscular dystrophy features in a mouse model (1), and are now trying it out in human cells. HACs, whether designed from scratch or built using natural human chromosomes as a framework, are slowly gaining in popularity. These artificial DNA molecules, which can exist in human cells as an extra, 47th chromosome, are useful in a variety of applications, from basic studies of gene and chromosome function to potential stem cell and, perhaps, gene therapy treatments. Eventually, some synthetic biologists would like to use HACs to hold entire man-made biological pathways. HACs can also complement the much-discussed CRISPR/Cas9 gene-editing tools, experts say. “CRISPR is good for creating specific and relatively small changes—deletions, insertions, mutations—in an existing genome,” says Alina Chan of HarvardMedical School in Boston. “HACs are for large, megabase-scale genome

Machine Learning and Citizen Science: Opportunities and Challenges of Human-Computer Interaction
Background and Aim: In processing large data, scientists have to perform the tedious task of analyzing hefty bulk of data. Machine learning techniques are a potential solution to this problem. In citizen science, human and artificial intelligence may be unified to facilitate this effort. Considering the ambiguities in machine performance and management of user-generated data, this paper aims to...
متن کاملThe Co-Constitution of Health Systems and Innovation; Comment on “What Health System Challenges Should Responsible Innovation in Health Address? Insights From an International Scoping Review”
Lehoux et al provide a timely and relevant turn on the broad and ongoing discussion around the introduction of health technology and innovation. More specifically, the authors suggest a demand-driven approach to health innovation that starts from identifying challenges and demands at the health system level. In this commentary, I review a number of underlying implications of their study in rela...
متن کاملFunctional Speed Reserve as a Proxy for the Anaerobic Speed Reserve Using the Critical Speed Concept
Background: Although maximal sprint speed (MSS) and the anaerobic speed reserve (ASR) provides valuable information about the speed profile of athletes, these parameters fall short of providing important information about sub-maximal metabolic thresholds. The only field test that can offer an estimate of a sub-maximal metabolic threshold is the 3-minute all-out test for running (3MT) which deli...
متن کاملApplying Psychophysiological Methods for Measuring User Experience: Possibilities, Challenges and Feasibility
Traditional methods to access the experience of a user such as subjective reports have certain disadvantages. Participants have to be asked for their experiences and emotions which interrupts the process of experience and flow. Psychophysiological methods offer data throughout the process of experience, which unfolds new possibilities for user experience (UX) evaluation. In this paper we provid...
متن کاملWhat Health System Challenges Should Responsible Innovation in Health Address? Insights From an International Scoping Review
Background While responsible innovation in health (RIH) suggests that health innovations could be purposefully designed to better support health systems, little is known about the system-level challenges that it should address. The goal of this paper is thus to document what is known about health systems’ demand for innovations. Methods We searched 8 databases to perfo...
